چکیده

Alternative splicing (AS) of messenger RNAs diversifies gene function by generating protein isoforms with different biological properties. Over 95% human genes are alternatively spliced, but the functional consequences most AS events still not known. Here, using long-read RNA sequencing, we identified 5,826 associated epidermal differentiation (percent spliced index >10%, FDR <0.01). were enriched in histone modification and phosphorylation (p < 0.001). The common type was cassette exon splicing, which an intervening between two other exons is included or skipped mRNA sequence. We functionally characterized a mitogen-activated kinase (MAP3K7), had been previously studied epidermis. Basal progenitors preferentially express shorter MAP3K7 isoform switch to longer suprabasal, differentiated epidermis inclusion additional 81 bp exon. Using isoform-specific knockdown overexpression, demonstrate that single necessary sufficient transition keratinocytes from replicating progenitor state high clonogenic capacity into post-mitotic state. also performed binding motif analysis sequences adjacent 26 proteins predicted mediate keratinocytes, including ten unstudied roles skin. In summary, our study defines repertoire alternative demonstrates potential this resource identify RBPs regulate differentiation.

The keratinocyte growth-differentiation switch.

Growth/differentiation control of normal epithelial cells has been relatively understudied, because of the complexities involved in their cultivation and characterization. The present review is focused on progress in this area using the mouse primary keratinocyte system. This system reproduces under well defined culture conditions the switch between epithelial cell growth and differentiation wh...

Regulation of apoptosis by alternative pre-mRNA splicing.

Apoptosis, a phenomenon that allows the regulated destruction and disposal of damaged or unwanted cells, is common to many cellular processes in multicellular organisms. In humans more than 200 proteins are involved in apoptosis, many of which are dysregulated or defective in human diseases including cancer. A large number of apoptotic factors are regulated via alternative splicing, a process t...

U1-independent pre-mRNA splicing contributes to the regulation of alternative splicing

U1 snRNP plays a crucial role in the 5' splice site recognition during splicing. Here we report the first example of naturally occurring U1-independent U2-type splicing in humans. The U1 components were not included in the pre-spliceosomal E complex formed on the human F1gamma (hF1gamma) intron 9 in vitro. Moreover, hF1gamma intron 9 was efficiently spliced even in U1-disrupted Xenopus oocytes ...

Regulation of Ich-1 pre-mRNA alternative splicing and apoptosis by mammalian splicing factors.

The importance of alternative splicing in regulating apoptosis has been suggested by findings of functionally antagonistic proteins generated by alternative splicing of several genes involved in apoptosis. Among these, Ich-1 (also named as caspase-2) encodes a member of the caspase family of proteases. Two forms of Ich-1 are produced as a result of alternative splicing: Ich-1L, which causes apo...
